Fly hit the nail on the head about the Overdrive control. As far as the tone knobs, the easiest way to go about it is to set them to where it sounds good to you. Sounds pretty unscientific, but it would be next to impossible for somebody to tell you where to set your amp controls over the internet without ever having heard you play.
